L-Tyrosine

Wat is L-Tyrosine?

L-tyrosine is een niet-essentieel aminozuur dat in het lichaam wordt gesynthetiseerd uit het essentiële aminozuur fenylalanine (7212). Uit voeding wordt het verkregen in bonen, noten, zaden, zuivelproducten, vlees, vis, eieren en sommige granen (7217). Aangezien de voedingsbehoeften van tyrosine afhankelijk zijn van fenylalanine, is tyrosine een essentieel aminozuur voor personen met Fenylketonurie (PKU). Deze personen zijn namelijk niet in staat om fenylalanine te consumeren in hun dieet (7212).

Oraal wordt tyrosine ingezet bij depressie, ADHD (Aandachtstekortstoornis), cognitieve prestaties en geheugen, fenylketonurie (PKU) en het verbeteren van de alertheid na slaapgebrek. Het wordt tevens gebruikt voor stress, premenstrueel syndroom (PMS), de ziekte van Parkinson, chronisch vermoeidheidssyndroom (CVS), narcolepsie, alcohol, heroïne en cocaïne-ontwenning, de ziekte van Alzheimer en andere dementievormen, hart- en vaatziekten, impotentie, verlies van libido, schizofrenie en als zonnebrandmiddel en eetlustremmer. Tyrosine wordt eveneens gebruikt om de trainingsprestaties te verbeteren en voor hypertensie. Het wordt ook gebruikt voor nemaline myopathie (NM) bij kinderen.

Plaatselijk wordt tyrosine gebruikt om tekenen van huidveroudering tegen te gaan, zoals bij rimpels.

Wetenschappelijke naam

2-amino-3-(4-hydroxyphenyl) propionic acid).

Wat is L-Tyrosine?

Werking

Tyrosine is een niet-essentieel aminozuur dat het lichaam synthetiseert uit fenylalanine. Voor mensen met fenylketonurie (PKU) die tyrosine niet kunnen synthetiseren uit fenylalanine, is tyrosine een essentieel aminozuur (7212). Tyrosine kan ook worden verkregen uit dieetproteïnen in zuivelproducten, vlees, vis, eieren, noten, bonen, haver en tarwe. Wanneer de inname van tyrosine via de voeding onvoldoende is, wordt fenylalanine omgezet in tyrosine (7217). De voedingsbehoeften van tyrosine zijn afhankelijk van de inname van fenylalanine. Wanneer de inname van fenylalanine voldoende is (ongeveer 9 mg / kg per dag), moet de tyrosine-inname via de voeding ongeveer 7 mg / kg per dag zijn (7214). Ontoereikende tyrosine-inname is zeldzaam in ontwikkelde landen. Tyrosine is opgenomen in alle eiwitten.

Antidepressieve effecten
Tyrosine kan een rol spelen bij depressie. Tyrosine is een voorloper voor de synthese van catecholamines, norepinefrine, epinefrine en dopamine (7215, 7217). Lage niveaus van norepinefrine kunnen een rol spelen bij depressie en dopamine kan een rol spelen bij de functie van antidepressiva (81470). Bij gezonde mensen verhoogt een acute inname van tyrosine de plasmaspiegels van noradrenaline, epinefrine en dopamine (81495). In menselijk onderzoek resulteerde het drinken van een aminozuurdrank die in de catecholamine-voorlopers tyrosine en fenylalanine ontbreekt in een verminderde reactie van de hersenen op prikkels; er waren echter geen functionele gedragsveranderingen (91465).

Antistress-effecten
Tyrosine is een voorloper van thyroxine en melanine (7212); het is ook een voorloper voor de synthese van catecholamines, norepinefrine, epinefrine en dopamine (7215, 7217). Er is interesse in tyrosine voor het voorkomen van de negatieve effecten gerelateerd aan stress. Sommige wetenschappers denken dat de hersenen onder stressvolle omstandigheden mogelijk onvoldoende tyrosine kunnen synthetiseren uit fenylalanine (7217). Catecholamines zoals epinefrine, noradrenaline en dopamine die worden gesynthetiseerd uit tyrosine kunnen tijdens stress uitgeput raken (81488). Er is speculatie dat het verhogen van de beschikbaarheid van tyrosine voor de hersenen een verhoogde catecholaminesynthese mogelijk maakt en negatieve effecten van stress vermijdt (91466, 91468). Er is enig bewijs bij dieren en mensen dat supplementaire tyrosine de prestaties, het geheugen en het leervermogen kan verbeteren, onder extreme omgevingsomstandigheden, intense lichaamsbeweging, psychologische stress (7215, 7217, 81476, 81484, 81501, 91470).

Werking

Veiligheid

Waarschijnlijk veilig

Wanneer oraal gebruikt in hoeveelheden die normaliter in voeding voorkomen. Tyrosine heeft de status Algemeen Herkend als Veilig (GRAS) in de Verenigde Staten (4912).

Wanneer oraal gebruikt en geschikt gebruikt in medicinale hoeveelheden, voor een korte termijn. Tyrosine is veilig gebruikt in doseringen tot 150 mg/kg dagelijks voor een periode van 3 maanden (7210, 7211, 7215). Wanneer plaatselijk en correct gebruikt (6155).

Zwangerschap en lactatie

Er is te weinig betrouwbare informatie beschikbaar over de veiligheid van tyrosine tijdens zwangerschap en in de lactatieperiode wanneer het gebruikt wordt in medicinale hoeveelheden. Sommige farmacokinetische onderzoeken tonen aan dat orale inname van 2-10 gram tyrosine oraal per dag de niveaus van vrije tyrosine in moedermelk bescheiden kan verhogen. De totale hoeveelheden worden hierbij echter niet beïnvloed. Daarom is het niet duidelijk of de toename van vrije tyrosine als zorg moet worden beschouwd (91467).

Effectiviteit

Effectief

Fenylketonurie (PKU)
Tyrosine wordt gebruikt als onderdeel van medische voedingsmiddelen voor mensen met fenylketonurie (PKU), een genetische aandoening waarbij de voorloper van tyrosine, fenylalanine, niet kan worden gemetaboliseerd (7212, 7213, 7232). De huidige richtlijnen bevelen aan dat mensen met PKU een dieet dat laag in fenylalanine aanhouden en de opname van tyrosine als onderdeel van medische voeding moeten handhaven om zo de normale tyrosine waarden in het bloed te behouden (95108). Suppletie met extra tyrosine wordt alleen aanbevolen voor patiënten met consistent lage hoeveelheden tyrosine in de bloedspiegels, ondanks het gebruik van tyrosine-bevattende medische voedingsmiddelen (95108). Routinesuppletie met gratis tyrosine wordt niet aanbevolen voor de meeste patiënten met PKU, omdat het grote variaties in plasmaconcentraties en bijwerkingen van tyrosine kan veroorzaken (7212, 95108). Tyrosine-inname verbetert de neuropsychologische maatregelen bij patiënten met PKU niet (81503, 91471, 95108).

Mogelijk effectief

Cognitieve prestaties
Uit voorlopig klinisch bewijs blijkt dat het nemen van 100 mg/kg tyrosine 2 uur voorafgaand aan cognitieve tests, de stemming, visuele reactietijd of auditieve reactietijd niet verbetert in vergelijking met placebo bij gezonde mannelijke personen (1143). Het gebruik van 100-300 mg/kg tyrosine voorafgaand aan acute blootstelling van stress, lijkt echter de door koude geïntroduceerde vermindering van cognitieve prestaties te verminderen in vergelijking met pacebo (81476, 81484). Bovendien lijkt het nemen van 100 mg/kg tyrosine voorafgaand aan cognitieve tests de uitvoerende werking en het korte termijngeheugen te verbeteren in vergelijking met placebo bij patiënten die werden getest tijdens door stress veroorzaakte stress (81501).

Geheugen
Het gebruik van tyrosine lijkt het geheugen te verbeteren onder stressvolle omstandigheden. Sommige klinische onderzoeken tonen bijvoorbeeld aan dat het nemen van 150-300 mg/kg tyrosine de geheugenprestaties verbetert wanneer het voorafgaand aan acute blootstelling van koude-stress wordt genomen (81477, 81499). Ook verbetert het nemen van 150 mg/kg een uur voorafgaand aan een multitasking-batterijentest het werkgeheugen tijdens een test vergelijken met een placebo (81469). Daarnaast verbetert het nemen van 150 mg/kg tyrosine 2 uur voorafgaand aan het uitvoeren van een geheugentaak het geheugen niet in vergelijking met placebo bij gezonde volwassenen die niet worden blootgesteld aan koude stress (81499).

Slaapgebrek
Orale inname van tyrosine lijkt de alertheid na slaapgebrek te verbeteren (7215). Tyrosine 150 mg / kg lijkt de prestatievermindering in psychomotorische tests ongeveer 3 uur na het verlies van een nachtrust uit te stellen (7215). Ander voorlopig klinisch onderzoek toont aan dat het nemen van tyrosine 150 mg / kg voorafgaand aan cognitieve testen het geheugen, redeneren en waakzaamheid verbetert in vergelijking met placebo bij slaaparme patiënten (81472).

Aandachtstekortstoornis met hyperactiviteit (ADHD)
Orale inname van tyrosine lijkt de symptomen van ADHD bij kinderen niet te verbeteren (7209). Bovendien lijkt het oraal innemen van tyrosine de symptomen van ADHD bij volwassenen niet te verbeteren, vooral door onoplettendheid. Sommige patiënten hebben mogelijk voorbijgaande verbetering van de symptomen na het starten van tyrosine, maar volwassen patiënten verwerven snel tolerantie (7210, 7211).

Depressie
Orale inname van tyrosine lijkt de symptomen van matige depressie niet te verbeteren (7208).

Trainingsprestaties
Het oraal nemen van tyrosine voorafgaand aan oefeningen op de loopband of op de fiets, uitgevoerd bij (matige) hitte, zorgt niet voor een toename van kracht, uithoudingsvermogen of prestaties in vergelijking met een placebo (81471, 81474, 81504, 91470).

Veiligheid

Bijwerkingen

Oraal, grote doseringen tyrosine 150 mg / kg dailcan kunnen misselijkheid, hoofdpijn, vermoeidheid, brandend maagzuur en gewrichtspijn veroorzaken (7211). Het dagelijks innemen van een combinatie van 4 gram tyrosine, 5 mg 5-hydroxytryptophan en 100 mg carbidopa kan diarree, misselijkheid, braken, slaperigheid en agitatie veroorzaken. Deze effecten kunnen worden beperkt door de dosering te verlagen (918).

Interacties

Medicijnen

Levodopa
Er is enige bezorgdheid dat tyrosine de effectiviteit van L-dopamine zou kunnen verminderen. Tyrosine en levodopa concurreren voor absorptie in de proximale twaalfvingerige darm door het grote neutrale aminozuur (LNAA) transportsysteem (2719). Adviseer patiënten om de dosering tyrosine en L-dopamine minimaal 2 uur te scheiden.

Schildklierhormoon
Er is enige bezorgdheid dat tyrosine additieve effecten kan hebben met schildklierhormoon medicijnen. Tyrosine is een voorloper van het schildklierhormoon en kan de niveaus verhogen (7212). Deze geneesmiddelen omvatten levothyroxine (Synthroid, Levoxyl) en liothyronine (Cytomel).

Lab testen

Schildklier stimulerend hormoon (TSH)
Theoretisch kan tyrosine TSH-niveaus verlagen. Tyrosine is een voorloper van thyroxine en kan schildklierhormoonspiegels verhogen, wat TSH-resultaten kan verminderen (7212).

Ziektes

Hyperthyreoïdie, ziekte van graves
Theoretisch kan tyrosine hyperthyreoïdie en de ziekte van Graves verergeren. Tyrosine is een voorloper van thyroxine en kan schildklierhormoonspiegels verhogen (7212). Vertel patiënten met hyperthyreoïdie of de ziekte van Graves om geen tyrosine supplementen te nemen.

Dosering

Tyrosine wordt bij volwassenen gebruikt in dagelijkse doseringen van 100-300 mg per kilogram lichaamsgewicht. Bij kinderen is onderzoek echter nog beperkt.
